| 目的通过研究高强度聚焦超声(high intensity focused ultrasound,HIFU)对裸鼠皮下人胶质瘤模型作用后的病理转归,探讨其治疗胶质瘤的有效性和安全性。方法选取24只裸鼠,建立裸鼠皮下人胶质瘤模型,随机分为对照组和实验组。用频率9.7MHz,焦距4.5mm,声强2500W/cm2的HIFU连续作用20s,于急性期(<3d)、亚急性期(4~14d)、慢性期(15~30d)观察各组肿瘤的生长情况;行2,3,5-三苯基氯化四氮唑(triphenyl tetrazoliumcholoride,TTC)染色、组织学切片及电镜检查。结果24只裸鼠全部成瘤,接种20d后肿瘤体积为(166.56±9.23)mm3,在HIFU作用7d后,实验组肿瘤体积倍增时间(tumor volume doubling time,TD)与对照组差异有统计学意义(P<0.05)。TTC染色显示胶质瘤的生物学焦域为(66.12±2.52)mm3。HE染色示照射区即刻就发生凝固性坏死,并随观察时间的延长坏死越明显;在亚急性期和慢性期可以观察到明显的交界区形成,且在照射区、交界区之间形成锐利的边界。电镜示细胞超微结构出现典... |
