糖皮质激素促β-淀粉样蛋白致海马神经元损伤

摘要:
目的探讨糖皮质激素(glucocoricoids,GCs)促进&beta;-淀粉样蛋白(&beta;-amyloid protein,A&beta;)引起海马神经元损伤的机制。方法地塞米松(dexamethasone,DEX)和&beta;-淀粉样蛋白与胎鼠海马神经细胞共同孵育后,用Western blot方法检测空白组、DEX组、A&beta;组、DEX+A&beta;组核因子&kappa;B(nuclear factor&kappa;B,NF-&kappa;B)、p-tau在细胞内的表达情况。结果与同剂量A&beta;组相比,DEX+A&beta;组的核NF-&kappa;B蛋白含量降低约49.2%,p-tau蛋白含量增加约150%,且差异有统计学意义(P<0.05)。结论在体外条件下GCs可能通过下调NF-&kappa;B蛋白水平、上调p-tau蛋白水平促进A&beta;引起海马神经元损伤。
