Teneurin蛋白羧基端相关肽的神经元保护作用

摘要：

目的研究肽类物质Teneurin蛋白羧基端相关肽(TCAP-1)对小鼠下丘脑神经细胞株Gn11增殖的影响。方法cAMP实验中,12孔培养皿中每孔加入3万Gn11细胞,每6孔为1组。细胞增殖实验中,96孔培养皿中每孔加入3千Gn11细胞,每8孔为1组。1,10,100nmol/LTCAP-1被分别加入不同实验组中。对照组中仅加入等量溶剂。通过酶标免疫法测定细胞内cAMP水平,通过MTT实验评估细胞增殖情况。在TCAP-1受体评估实验中,各实验组(n=6)中分别加入1nmol/LTCAP-1,1nmol/LTCAP-1+100nmol/LPD171729,1nmol/Lurocortin,1nmol/Lurocortin+100nmol/LPD171729和100nmol/LPD171729。对照组中只加入溶剂。观察Gn11细胞cAMP改变。结果在cAMP实验中,TCAP-1作用15min后,低浓度(1nmol/L)TCAP-1导致细胞cAMP水平明显升高(P<0.05),高浓度(100nmol/L)TCAP-1导致细胞cAMP水平明显降低(P<0.05)。TCAP-1的作用与CRFR1受体无明显相关性。在MTT...
